The Russo brothers, known for their work on previous Avengers films, are back in charge, and Robert Downey Jr. is returning to the Marvel Cinematic Universe (MCU) as Doctor Doom, the big villain of this movie.
Has Marvel confirmed ‘Avengers: Doomsday’s first characters?

So far, we have six confirmed characters for Doomsday: Doctor Doom, the Fantastic Four (Reed Richards, Sue Storm, Johnny Storm, and Ben Grimm), and Doctor Strange, who was previously hinted at by Benedict Cumberbatch. While it’s likely we’ll see other characters like Deadpool and Wolverine, the Fantastic Four are clearly central to the story.
